dc.description.abstractgerÜber die Existenz des neurenterischen Kanals und seinen potentiellen Einfluss auf fetale Malformationen wie neuroendodermale Zysten, junctional neurulational defects und dem split notochord syndrome diskutieren Embryologen und klinisch tätige Ärzte seit mehr als hundert Jahren. Durch die Seltenheit von menschlichen Embryonen der fraglichen Entwicklungszeit von neunzehn bis einundzwanzig Entwicklungstagen (Carnegie-Stadien 8 und 9) und ihre weltweite Verteilung war eine fundierte vergleichende Untersuchung stets erschwert. Der in dieser Arbeit erfolgte, systematische und kleinschrittige Vergleich der Histologie von fünf menschlichen und zwei Weißbüschelaffenembryonen der Stadien 8 und 9 sowie deren partielle virtuelle Rekonstruktion ermöglichen erstmals die Beschreibung des neurenterischen Kanals als physiologische Struktur der Embryogenese von Primaten. Als senkrechter Kanal im Bereich des Primitivknotens verbindet er Amnionhöhle mit Dottersack und ermöglicht eine hufeisenförmige Involution des Ektoderms. Hierdurch ensteht die Chordaplatte, die wiederum die Neurulation induziert. Darüber hinaus wiesen die asymmetrische Expression des Gens Nodal im paraaxialen Mesoderm eines Embryos und morphologische Asymmetrien in sechs von sieben untersuchten Embryonen auf eine frühe, wahrscheinlich von Zilien unabhängige Etablierung der Links-/Rechtskörperachse bei Primaten hin. Daraus lässt sich schließen, dass der neurenterische Kanal der Primaten eine funktionelle und morphologische Homologie des in Amphibien, Reptilien und einigen Vögeln beschriebenen Blastoporus darstellt. Damit wären bei Primaten zwei bisher nur separat betrachte Prozesse der Gastrulation, die Involution an einem Kanal und die Ingression entlang einer Achse, in einem Organismus nachweisbar. Der Prozess der Gastrulation, also die Anlage der drei Keimscheibenblätter, wäre damit komplexer als bisher angenommen. Die räumlichen Zusammenhänge dieser schwierig zu verstehenden, mikroskopisch kleinen Strukturen konnten dabei durch die Anwendung von nachvollziehbaren Methoden der digitalen Rekonstruktion und des anschließenden dreidimensionalen Drucks „begreifbar“ dargestellt werden. Gerade für die Ausbildung von Studenten und jungen Ärzten bietet sich hierdurch die Chance, klassische embryologische Lehre durch neue, interaktive Konzepte zu unterstützen.de
dc.description.abstractengThe existence of the neurenteric canal and its potential influence on fetal malformations such as neuroendodermal cysts, junctional neurulational defects and split notochord syndrome has been discussed by embryologists and clinicians for more than a hundred years. Due to the rarity of human embryos of the developmental period in question of nineteen to twenty-one developmental days (Carnegie stages 8 and 9) and their worldwide distribution, a well-founded comparative study has always been difficult. The systematic, small-step comparison of the histology of five human and two common marmoset monkey embryos of stages 8 and 9 as well as their partial virtual reconstruction in this study allows for the first time to describe the neurenteric canal as the physiological structure of primate embryogenesis. As a vertical canal in the area of the primitive node, it connects amniotic cavity with yolk sac and enables a horseshoe-shaped involution of the ectoderm. This creates the notochordal plate, which induces neurulation. In addition, the asymmetric expression of the gene nodal in the paraxial mesoderm of an embryo and morphological asymmetries in six of seven embryos studied indicate an early establishment of the left/right body axis in primates, probably independent of cilia. From this it can be concluded that the neurenteric canal in primates represents a functional and morphological homology of the blastoporus described in amphibians, reptiles and some birds. Thus, two processes of gastrulation in primates, involution along a canal and ingression along an axis, which have so far only been considered separately, could be detected in one organism. The process of gastrulation, i.e. the formation of the three germinal disc layers, would thus be more complex than previously assumed. The spatial relationships of these microscopically small structures, which are difficult to understand, could be "grasped" by using comprehensible methods of digital reconstruction and subsequent three-dimensional printing. Especially for the training of students and young doctors this offers the chance to support classical embryological teaching with new, interactive concepts.de
