dc.description.abstractengBone regeneration of critical-size defects requires ideal release on time of growth factors. The aim of the present in vivo study on male Wistar rats was to improve bone regeneration in mandibular defects of critical-size through the combined release of osteogenic (rhBMP-2) and angiogenic (rhVEGF) factors. It was a matter to find out how the combination of VEGF and BMP could improve bone regeneration and to reduce the dose of BMP. The polylactide/calcium carbonate composite scaffolds were loaded in single-doses and combined doses with 24 µg, 48 µg, and 96 µg BMP as well as 1.5 µg and 6 µg VEGF by dropping and adsorption. The resulting implants could be inserted into the mandibular defects. The observation periods were set at 4 weeks and 13 weeks. After the observation time and the preparation of the jaws, we carried out the radiological examination using DVT and subsequent computer-aided radiomorphometric evaluation on reconstructed axial images. In a comparison of the combination groups with the single-doses of the BMP, the bone density and bone area of the 96 µg BMP single-dose group showed a comparable increase over the observation period as the combination groups with 48 µg BMP. The results of this study can contribute to a better understanding of the interaction of the growth factors. The effect of increasing bone tissue regeneration, in which the BMP concentration can be halved by the addition of VEGF, was shown in this study.de
