dc.description.abstractgerDie Laminin-Untereinheiten α1, α3, α5, β1, und γ1 wurden in dieser Arbeit als Bestandteile der extrazellulären Matrix des menschlichen Knorpels und in den mesenchymalen Extremitätenanlagen nachgewiesen, wobei von allen, bis auf Laminin-α1, noch nicht bekannt war, dass sie in diesen überhaupt vorkommen. Generell konnte nachgewiesen werden, dass mehr Laminin im osteoarthritischen Knorpel vorkommt als im gesunden. Dabei fiel auf, dass Männer mehr Laminin-α3 exprimieren als Frauen, welche bekanntlich häufiger an Osteoarthritis erkranken als Männer. Es konnte gezeigt werden, dass die Menge von Laminin um die elongierten Chondrozyten bzw. die chondrogenen Progenitorzellen aus den späten Stadien der Osteoarthritis am stärksten erhöht war. Dies weist auf deren Regenerationsaktivität hin. Ebenfalls konnte gezeigt werden, dass eine Lamininstimulation von osteoarthritischen Chondrozyten eine verstärke Expression von Bestandteilen der extrazellulären Matrix, welche gesunden Verhältnissen entspricht, hervorruft. Auch dies lässt auf eine Bedeutung von Laminin-Ketten für das regenerative Potential der chondrogenen Progenitorzellen schließen. Zum Abschluss wurden adhäsive Eigenschaften spezifisch für Chondrozyten aus den späten Stadien der Osteoarthritis gezeigt, und Chondrozyten-Laminin-Interaktionen genauer beleuchtet. Insgesammt lassen die Ergebnisse den Schluss zu, dass die untersuchten Laminine den Chondrozyten und den chondrogenen Progenitorzellen dabei helfen, mögliche Regenerations-prozesse durchzuführen und die Knorpelintegrität in den späten Stadien der Osteoarthritis wiederherzustellen. Deshalb scheint es möglich, dass diese eine Schlüsselrolle in der Pathogenese der Osteoarthritis spielen.de
dc.description.abstractengThe laminin subunits α1, α3, α5, β1, and γ1 were detected in this study as components of the extracellular matrix of human cartilage and in the mesenchymal limb systems, and by all, except for laminin-α1, was not known that they occur in this at all. Generally it was found that more laminin is present in osteoarthritic cartilage than in the healthy. It was noticeable that men express more laminin α3 as women who suffer from osteoarthritis than men known to frequent.
