dc.description.abstractgerNach wie vor gibt es im Bereich der neurologischen Erkrankungen viele ungelöste Fragen und Probleme. Zu diesen gehören die Synucleinopathien, bei denen die Akkumulation und Aggregation von α-Synuclein pathogenetisch im Vordergrund steht. Diese Arbeit zielt darauf ab, α-Synuclein mit Hilfe des Nanobodies NBSyn87 zu detektieren. Hierzu wird zunächst gezeigt, dass der Nanobody in der Lage ist, mit hoher Spezifität an menschliches α-Synuclein zu binden. Darüber hinaus wurde der Nanobody - eingebunden in ein Tet-on-3G-System - in humanen HEK-Zellen eingesetzt, um intrazelluläres α-Synuclein nachzuweisen. Hierzu wurde der Nanobody an ein GFP-Signal gekoppelt, sodass die qualitative und quantitative Auswertung mit Hilfe eines Fluoreszenz-Mikroskopes ermöglicht wird.  Ziel ist es, mit Hilfe des Nanobodys in Zukunft Aufschlüsse über die Transmission und Aggregation von α-Synuclein und somit wichtige Informationen über die Pathogenese der Synucleinpathien gewinnen zu können. de
dc.description.abstractengThere are still many unresolved issues and problems in the field of neurological diseases. These include synucleinopathies. Accumulation and aggregation of α-synuclein are pathogenetic hallmarks for synucleinopathies. This work aims to detect α-synuclein using the nanobody NBSyn87. For this purpose, it is first shown that the nanobody is able to bind with high specificity to human α-synuclein. In addition, the nanobody - integrated into a tet-on-3G-system - was used in human HEK cells to detect intracellular α-synuclein. For this purpose, the nanobody was coupled to a GFP-signal, thus it is possible to evaluate the results by fluorescence microscopy.  The aim is to be able to obtain information about the transmission and aggregation of α-synuclein and thus important information about the pathogenesis of the synucleinpathies with the help of the nanobody in the future. de
