dc.description.abstractgerDas zelluläre Prionprotein ist ein multifunktionales, evolutionär hochgradig konserviertes und ubiquitär exprimiertes Protein. Es durchläuft eine Reihe von Modifikationen während seiner posttranslationalen Prozessierung, wobei verschiedene Glykoformen und trunkierte Fragmente resultieren. Es existieren nur wenige Daten über die Expression und Spaltung des zellulären Prionproteins beim Menschen. In dieser Studie untersuchten wir die Zusammensetzung von Prionprotein-Isoformen im Liquor cerebrospinalis von Patienten mit verschiedenen humanen Prionenerkrankungen. Die erste Gruppe von Patienten war von einer sporadischen Creutzfeldt-Jakob-Krankheit betroffen, bei der verschiedene Codon-129-Genotypen zu finden sind. Die zweite Gruppe umfasste Patienten mit einer genetischen Form der Creutzfeldt-Jakob-Krankheit, bei denen eine E200K-Mutation vorliegt. Die dritte Gruppe repräsentierte Patienten mit einer letalen familiären Insomnie, und die letzte Gruppe bestand aus Patienten mit einem Gerstmann-Sträussler-Scheinker-Syndrom. Wir untersuchten, ob der Codon-129-Polymorphismus in der sporadischen Creutzfeldt-Jakob- Krankheit und ebenso der Typ der Prionenerkrankung in Menschen Auswirkungen auf die Glykosylierung und Prozessierung des zellulären Prionproteins hat. Immunoblot-Analysen unter Verwendung verschiedener monoklonaler Prionprotein-Antikörper, die gegen unterschiedliche Epitope des zellulären Prionproteins gerichtet sind, zeigten für alle untersuchten Gruppen von Patienten ein einheitliches Vorherrschen der glykosylierten Prionprotein-Isoformen im Vergleich zur unglykosylierten Form. Darüber hinaus detektierte der Antikörper SAF70 eine Reihe von Prionprotein-Fragmenten der Größe 21, 18, 13 und 12 kDa. Unsere weiteren experimentellen Funde deuteten darauf hin, dass die Polymorphismen am Codon-129 des Prionprotein-Gens, die E200K-Mutation und die verschiedenen untersuchten Typen humaner transmissibler spongiformer Enzephalopathien keinen messbaren Einfluss auf die Glykosylierung und Prozessierung des zellulären Prionproteins bei humanen Prionenerkrankungen ausüben.de
dc.description.abstractengThe cellular prion protein is a multifunctional, evolutionarily highly conserved and ubiquitously expressed protein. It goes through a number of modifications during its post-translational processing, resulting in various glycoforms and truncated fragments. Little data exist on the expression and cleavage of the cellular prion protein in humans. In this study, we investigated the composition of prion protein isoforms in the cerebrospinal fluid of patients with various human prion diseases. The first group of patients was affected by a sporadic Creutzfeldt-Jakob disease in which different codon 129 genotypes can be found. The second group included patients with a genetic form of Creutzfeldt-Jakob disease who had an E200K mutation. The third group represented patients with fatal familial insomnia and the last group consisted of patients with Gerstmann-Sträussler-Scheinker syndrome. We investigated whether the codon 129 polymorphism in the sporadic Creutzfeldt-Jakob disease and also the type of prion disease in humans have effects on the glycosylation and processing of the cellular prion protein. Immunoblot analyzes using various monoclonal prion protein antibodies which are directed against different epitopes of the cellular prion protein showed a uniform predominance of the glycosylated prion protein isoforms compared to the unglycosylated form for all groups of patients examined. In addition, the SAF70 antibody detected a number of prion protein fragments measuring 21, 18, 13 and 12 kDa. Our further experimental findings indicated that the polymorphisms at codon 129 of the prion protein gene, the E200K mutation and the various types of human transmissible spongiform encephalopathies investigated have no measurable influence on the glycosylation and processing of the cellular prion protein in human prion diseases.de
