dc.description.abstract | In dieser Arbeit wurden drei bZIP-Transkriptionsfaktoren, BZI-2, BZI-3 und BZI-4 isoliert und charakterisiert, die mit BZI-1 heterodimerisieren. Im Gegensatz zu BZI-1, das ubiquitär exprimiert wird, zeigen die anderen BZI-Proteine differentielle Expressionsmuster. BZI-2 wird vor allem in den Sink-Organen wie der Blüte, den Wurzeln, aber auch im Stengel und in jungen Blättern exprimiert. Die Transkriptmenge wird durch Licht und Zucker induziert. Die BZI-3 RNA wird vor allem in der Blüte transkribiert, liegt aber auch in allen anderen Geweben vor. Durch einen Kältestimulus steigt die Transkriptmenge an. BZI-4 wird in der Blüte exprimiert, in den Stamina ist die Expression von BZI-4 am höchsten. Die Überexpression von BZI-3 und BZI-4 führt zu pleiotropen Veränderungen der Pflanze. BZI-3 Überexpression führt zu einer Reduktion der durch Auxin hervorgerufenen Zellstreckung und Wurzelregeneration. Die BZI-4 überexprimierende Pflanze weist verkürzte Internodien auf. Auch die Blütenorgane beider Linien sind verkürzt. Alle BZI-Proteine binden in vitro an den Promotor des Auxinregulierten GH3-Gens. Für BZI-1 konnte diese Bindung auch in planta durch Chromatin-Immunopräzipitation bestätigt werden. In dieser Arbeit konnte die selektive Bindung von BZI-4 Homodimeren an ein Promotorelement einer antherenspezifischen Invertase, der NIN88 gezeigt werden. Die Expression der NIN88 ist in BZI-4-Oex Pflanzen reduziert. | de |

dc.description.abstracteng | Three bZIP-transcription factors, BZI-2, BZ-3 and BZI-4 were isolated, which heterodimerise with BZI-1. In contrast to BZI-1, these proteins show distinct expression patterns. BZI-2 is expressed in flowers and roots and also in the stem and young leaves. The RNA-levels are induced by sugars and light. BZI-3 is mainly expressed in the flowers, but is also represented in all other tissues. After exposure to cold, the transcript level is enhanced. BZI-4 is expressed in the flowers, it is most abundant in the anthers. Overexpression of BZI-3 and BZI-4 leads to pleiotropic phenotypes. Overexpression of BZI-3 leads to a reduction in auxin dependent cell elongation and root regeneration. Overexpression of BZI-4 leads to shortened internodes. The flower organs of both lines are shortened. The BZI proteins are binding in vitro to the promoter of the GH3 gene. By using chromatin immunoprecipitation, a binding of BZI-1 to the GH3 promoter could be proven in planta. The selective binding of BZI-4 homodimers to a promotor element of the NIN88 gene coding for an anther specific invertase could be shown.
