dc.description.abstract | Geminin reguliert DNA Re-Replikation während eines
Zellzyklus durch Interaktion mit dem
Lizenzierungs-Faktor Cdt1. Darüber hinaus bindet es an
mehrere Transkriptionsfaktoren, darunter
Homeodomain-Proteine, wie Hox und Six3, und Mitglieder
von Chromatin-Remodelling-Komplexen, insbesondere von
Repressoren der Polycomb Gruppe, und von Bestandteilen
des SWI/SNF Komplexes. Hier beschreibe ich die
biochemische Analyse des bisher uncharakterisierten
Proteins TIPT (TATA-binding protein-like
factor-interacting protein), das im Two-Hybrid Screen
als starker Bindungspartner des murinen
Geminin-Proteins identifiziert wurde. TIPT wird in
embryonalen und adulten Geweben der Maus exprimiert,
sowie in Gewebekultur-Zellen, wo es sich im Zytoplasma,
in Centrosomen und im Nukleolus befindet. Wie Geminin,
interagiert auch TIPT mit Mitgliedern der Polycomb
Gruppe, einschließlich Sex comb on midleg homolog
(Scmh1), und darüber hinaus mit dem Transkription
Faktor AF10, sowie mit nukleolären und centrosomalen
Proteinen. Geminin und TIPT interagieren mit dem
allgemeinen Transkriptionsfaktor TBP (TATA box binding
protein), und mit dem TBP-like factor (TBPL1/TRF2).
TIPT Überexpression in Kultur-Zellen verlängert die S
Phase und verkürzt die G2/M Phase des Zellzyklus, ohne
die Zellproliferation zu beenflussen, wie nach stabile
TIPT-Transfektion zu beobachten ist. Die nukleoläre
Lokalisation von TIPT ist verändert nach
Herunterregulation der Pol I-abhängigen Transkription,
oder nach RNA Depletion. Die Überexpression von TIPT
aktiviert die endogene Pol I-Transkription und die
Transkription von Pol I-Reportern. TIPT bindet in vitro
den Adenovirus-major-late-Promoter an einem BRE Element
vor der TATA Box und bildet einen ternären Komplex mit
TBP. Ein ähnlicher Komplex bildet sich mit TBPL1 und
dem Neurofibromin (NF1) Promoter. TIPT bewirkt zusammen
mit Geminin und TBP die Aktivierung des TATA
Box-Promoters, und mit TBPL1 und Geminin die
Aktivierung des TATA-freien NF1 Promoters. Zusammen
gefaßt beschreibt diese Dissertation einen neuartigen ,
weit verbreiteten, transkriptionellen Ko-Aktivator und
seine Funktion in Zusammenarbeit mit
Chromatin-assoziierten Faktoren und der basalen
Transkriptionsmaschinerie. | de |

dc.description.abstracteng | Geminin prevents DNA re-replication during one cell
cycle by interacting with the licensing factor Cdt1. In
addition, it binds to several transcription factors
including Hox and Six3 homeodomain proteins, and to
members of the chromatin remodeling complexes, in
particular the polycomb group of repressors and the
SWI/SNF complex. Here I describe the biochemical
analysis of an uncharacterized protein TIPT
(TATA-binding protein-like factor-interacting protein),
identified in a two-hybrid screen as strong binding
partner of murine geminin. TIPT is widely expressed in
mouse embryonic and adult tissues, as well as in tissue
culture cells where is found in the cytoplasm, the
centrosome and the nucleolus. Like geminin, TIPT also
interacts with polycomb group members including Sex
comb on midleg homolog (Scmh1), and in addition with
AF10 transcription factor, as well as with nucleolar
and centrosomal proteins. Both geminin and TIPT
interact with the general transcription factor TBP
(TATA box binding protein), and with TBP-like factor
(TBPL1/TRF2). TIPT overexpression in culture cells
elongates S phase and shortens G2/M phase of the cell
cycle, without affecting cell proliferation observed
after stable TIPT transfection. TIPT nucleolar
localization is perturbed after Pol I drug-dependent
transcription inhibition, or after RNA depletion. TIPT
overexpression activates endogenous and reporter Pol I
transcription. TIPT binds in vitro the adenovirus major
late promoter on the BRE element upstream of the TATA
box and forms a ternary complex with TBP. It forms a
similar complex with TBPL1 and the neurofibromin (NF1)
promoter. TIPT synergizes with geminin and TBP in the
activation of TATA box-containing promoters, and with
TBPL1 and geminin in the activation of TATA-less NF1
promoter. Together, this doctoral dissertation
introduces a novel, widely expressed transcriptional
co-activator and its function in cooperation with
chromatin associated factors and the! basal
