dc.description.abstract | Verticillium longisporum ist ein bodenbürtiger Pilz,
welcher die Rapswelke hervorrufen kann. Das Pathogen
dringt über die Wurzeln in die Wirtspflanze ein und
wächst in Form eines Myzels im Xylem. Im späteren
Infektionsverlauf befällt es schließlich die übrigen
Gewebe der Pflanze. Um die Reaktionen infizierter
Pflanzen zu charakterisieren wurde die Modelpflanze
Arabidopsis thaliana gewählt. Es wurden
Transkriptomprofile der Rosetten fünf und 18 Tage nach
Infektion (dpi) aufgenommen. Fünf dpi zeigt A. thaliana
noch keine Symptome. Es werden jedoch eine Reihe von
Genen mit Bezug zur Jasmonsäure (JA) biosynthese oder
singnaltransduktion induziert. Durch die Infektion von
Pflanzen mit einer Mutation im JA-Biosyntheseweg wurde
der Einfluss dieser Gene auf die Symptomentwicklung bei
Arabidopsis untersucht. Die Reaktion der Pflanze 5 dpi
beeinflusste den Pathophenotyp und die Expression der
Verticillium longisporum induzierten Gene (VliGs) 18
Tage nach Infektion nicht. Mehr als ein Viertel (5 dpi)
bzw. ein Drittel (18 dpi) der VliGs codiert für
apoplastisch lokalisierte Gene, welche an
Zellwandmodifikationen und potentiellen
Abwehrreaktionen beteiligt sind. Ihre Induktion war

dc.description.abstracteng | Verticillium longisporum is a soil-borne fungal
pathogen causing vascular disease predominantly in
oilseed rape. The pathogen enters its host through the
roots and maintains a parasitic life stage in the xylem
before invading other tissues late in the infection
cycle. Arabidopsis thaliana was used as a model plant
to characterize the response of the aerial parts of the
plant to the infection. Whole genome transcriptome
profiling experiments of Arabidopsis Col-0 plants were
performed at 5 and 18 dpi, respectively. At 5 dpi, no
symptoms were detected, but a number of genes related
to jasmonic acid (JA) biosynthesis or signal
transduction were already expressed. However, this
initial defense reaction did not influence the
pathophenotype and the expression of Verticillium
induced genes (VliGs) at 18 dpi as revealed by analysis
of an Arabidopsis mutant deficient in JA biosynthesis.
More than one fourth (5 dpi) and one third (18 dpi) of
the VliGs encode apoplastically localized proteins
involved in cell wall modifications and potential
defense responses. Their expression was dependent on
the specific fungal isolate. The identified VliGs
provide a useful tool to elucidate the contribution of
the induced genes to the disease phenotype and the
defense response. Moreover, they might help to identify
the elicitor(s) as suggested by preliminary
experiments. They demonstrated that fractionated
extracts from fungal cultures induced expression of a
VliG when applied to the petioles of detached
