dc.description.abstractengThis dissertation focuses on the effects of receptor-dimerization regarding the muscarinic Acetylcholine-receptors M1, M3 and M5. The goal was to evaluate whether the dimerization between homo- or heterologous receptor partners between M1, M3, and M5 have an impact on the following intracellular receptor-signalling. We developed a method to enforce dimerization between two recepor-partners by generating receptors which have attached either an antigen (GFP) or an anti-GFP-antibody. By co-expressing both modified receptors on the cell surface we could ensure a forced dimerization between the two receptor partners due to direct antibody-antigen binding. We interpreted the impact of dimerization by measuring and comparing the intracellular calcium-levels in with live cell calcium-imaging. The results showed, that dimerization between two mAChRs can lead to potential coupling mechanism between the receptors and have an impact on the signalling of those.de
