dc.description.abstractengThe aim of this in vitro study was to investigate the release kinetics of rh-BMP-2 from polyelectrolyte multilayers on titanium platelets of different modifications. The heparin PEM showed sufficient loading of rh-BMP-2 and comparable release amounts and kinetics as previous studies (Guillot et al. 2013). After adding collagen to the coating to better simulate the ECM, a slightly lower amount of released rh-BMP-2 was shown, but with a release amount of at least 100 ng/72 h over 21 days, which seems necessary to induce efficient bone neosynthesis (Gruber et al. 2009). The cross-linking of the polyelectrolyte multilayers did not lead to the hoped-for delayed release kinetics. It was found that chemical crosslinking by EDC/NHS reaction led to a comparable release in terms of amount of protein and release pattern as the non-crosslinked coatings, whereas photochemical crosslinking by UV-crosslinking led to a very low, unusable BMP-2 release. de
