Vegetally localized mRNA molecules in Xenopus laevis oocytes play important roles in the formation of the primary germ layers and the germ line. In order to identify novel vegetally localized RNA molecules, 10000 cDNA clones of a library enriched with cortically and vegetally localized transcripts were analyzed by macro- and microarray hybridisations. By verification with whole mount in situ hybridisations 55 novel vegetally localized transcripts were identified. These novel transcripts were grouped into the major localization pathways according to their mRNA distribution pattern and further classified into functional groups. One of the novel identified and late localizing clones corresponds to the Xenopus homologue of a putative RNA binding protein Dead end and is expressed in the germ plasm and primordial germ cells during early Xenopus embryogenesis. The inhibition of XDead end protein translation by morpholino oligonukleotid injections results in the failure of primordial germ cells to migrate and suggest a conserved role during germ cell development. The cis-acting element, which mediates the transport to the vegetal pol, has been mapped to a 251 nt comprising region in the 3`-UTR of the XDead end RNA. This RNA localization element shows protein binding pattern characteristic for the late localization pathway involving one additional 38 kDa protein.

Am vegetalen Pol der Xenopus laevis lokalisierte mRNA Moleküle haben eine wichtige Funktion während der frühembryonalen Entwicklung. Mit dem Ziel, neue vegetal lokalisierte mRNA Moleküle zu isolieren, wurden 10000 cDNA Klone einer mit vegetal-kortikal lokalisierten Transkripten angereicherten cDNA Bibliothek mit Hilfe von Macro- und Microarray Hybridisierungen durchmustert. Dabei konnten 55 neue vegetal lokalisierte mRNA Moleküle identifiziert werden, die sich ausgehend vom räumlichen und zeitlichen Expressionsmuster in die Guppe der früh, spät oder intermediär lokalisierte RNAs einteilen lassen und außerdem interessante Funktionsgruppen bilden. Eine der neu identifizierten und über den späten Lokalisationsweg transportierten RNA kodiert für das Xenopus Dead end Homolog einhypothetischen RNA-Bindeprotein und ist im Keimplasma und in den Keimzellvorläuferzellen während der frühen Xenopus Embryogenese exprimiert. Die Unterdrückung der XDead end Proteintranslation druch Morpholino Oligonukleotid Injektionen resultiert im Ausbleiben einer korrekten Migration der Keimzellvorläuferzellen und deutet auf eine konservierte Funktion XDead ends innerhalb der Keimzellentwicklung hin. In Mikroinjektionsexperimenten konnte ein 251 Nukleotide umfassendes Lokalisationselement identifiziert werden, das für den Transport der XDead end RNA an den vegetalen Pol der Oozyte verantwortlich ist. Dieses Fragment zeigt zusätzlich zur Identifikation eines spezifisch an das XDead end Lokalisationselement bindendes Protein ein für spät lokalisierte RNAs charakteristisches Proteinbindungsmuster.
