dc.description.abstract | Das as-1-Element, welches ursprünglich aus dem Blumenkohl-Mosaik-Virus isoliert wurde, ist induzierbar durch Salizylsäure (SA), Auxin und Pathogenbefall. Elemente, die dem as-1-Element ähnlich sind, wurden in Promotoren vieler Pathogenabwehr-Gene gefunden. Zu ihnen zählen z.B. das früh exprimierte Gen Nt103 eine Glutathion-S-Transferase oder das spät exprimierte Gen PR1a. DNA-Bindestudien (EMSA) zeigten, dass das as-1-Element von den bZIP-Transkriptionsfaktoren TGA2.1 und TGA2.2 erkannt wird. Um eine in vivo-Interaktion der TGA-Faktoren mit endogenen as-1-ähnlichen Promotorelementen zu untersuchen, wurde die Chromatin-Immunopräzipitation (ChIP) für Tabak entwickelt. Die Etablierung erfolgte über die Verwendung eines Tetrazyclin-induzierbaren Promotorsystems, bei welchem zwei unterschiedliche Bindungszustände eines artifiziellen Transkriptionsfaktors untersucht werden konnten. ChIP-Experimente mit Pflanzen, die das Reportergenkonstrukt as-1::GUS tragen, zeigten eine konstitutive Bindung des TGA2.2 am as-1-Element nach Salizylsäure-Stimulus, während für PR1a eine induzierbare Bindung nachgewiesen werden konnte. | de |

dc.description.abstracteng | The as-1 element, originally found in the Cauliflower Mosaic virus, is inducible by salycylic acid, auxin and also by pathogen attack. Several as-1-like elements were also found in plant promoters of pathogen defense genes like the early response gene Nt103 (a glutathione-S-transferase) or the late responsive pathogenesis related protein PR1a. Binding studies using EMSA revealed recognition of the as-1 element by the TGA factors TGA2.1 and TGA2.2, members of the bZIP transcription factor family. To investigate the in vivo interaction between the TGA factors and as-1 like promoter elements a chromatin immunoprecipitation assay (ChIP) for tobacco leaves was developed. Establishment of the technique was proofed using the tetracyclin-inducible expression system in tobacco plants were two different conditions were tested successfully.
