dc.description.abstract | Die vorliegende Dissertation ist in zwei Teile gegliedert. Der erste Teil beschäftigt sich mit der Totalsynthese der Mansouramycine A-D und zeigt bisher gegangene Syntheseansätze zur Synthese von Mansouramycin E. Der zweite Teil handelt über eine neue, hoch-enantioselektive Rhodium-katalysierte 1,2-Addition von Aluminiumorganylen an cyclische Enone und En-1,4-dione. Die Mansouramycine wurden in der Gruppe von Prof. Dr. Laatsch in Göttingen isoliert. Erste Tests zeigten eine moderate biologische Aktivität gegen Staphylococcus aureus, Bacillus subtilis und Escherichia coli. Weitere Untersuchungen ergaben zudem eine vielversprechende Cytotoxizität gegen verschiedene menschliche Tumorzelllinien. Mansouramycin D wirkt beispielsweise mit einem durchschnittlichen IC50-Wert von 0.089 μM gegen Lungen- und Blasenkrebs, weshalb eine Totalsynthese sinnvoll erschien. Ausgehend von einem 2,5-Dimethoxybenzaldehyd wurde über eine Henry-Reaktion und anschließende Reduktion das 2,5-Dimethoxybenzylaminderivat gewonnen, welches nach Cyclisierung über eine Pictet-Spengler-Reaktion und Oxidation an Palladium zum entsprechenden Isochinolin umgesetzt wurde. Nach Oxidation an CAN konnte das Chinolindion-Derivat erhalten werden, welches mit dem entsprechenden Alkylamin selektiv aminiert wurde, so dass die Mansouramycine A-D in größeren Mengen mit einer Gesamtausbeute von 7-32% synthetisiert werden konnten. Eine hoch-enantioselektive Rhodium-katalysierte 1,2-Addition von Aluminiumorganylen an cyclische Enone wurde erstmals in der Arbeitsgruppe von v. Zezschwitz an der Universität Marburg entwickelt. In dieser Dissertation wurden u.a. cyclische En-1,4-dione an [Rh(cod)OMe]2 mit (R)-BINAP durch Trimethylaluminium selektiv an der 2-Postion alkyliert. Bei entsprechender Schutzgruppenchemie konnten beide Zentren mit einem ee bis zu 98% alkyliert und so der tertiäre Alkohol gewonnen werden, eine 1,4-Addition blieb dabei vollständig aus. Ausbeuten varierten nach Substraten von 40-90%. | de |

dc.description.abstracteng | The following thesis is divided into two sections. The first part deals with the total synthesis of Mansouramycin A-E. The second part shows a new high enantioselective rhodium catalyzed 1,2-addition of aluminum-organyles to cyclic enones and en-1,4-diones. The Mansouramycines where isolated in the Laatsch group at the department of Chemistry in Göttingen. They show a high cytotoxicity against many human cancer cell lines with an IC50 up to 0.089 μM for lung cancer. The total synthesis started with a Henry reaction of a 2,5-dimethoxybenzaldehyde to give after reduction the corresponding amine. This was cyclized by a Pictet-Spengler reaction and oxidized at palladium to give the isochinoline. After oxidation with CAN the resulting chinolindione was selectively aminated to give the corresponding Mansouramycine in a total yield of 7-32%. A high enantioselektive rhodium catalyzed 1,2-addition of aluminum organyles to cyclic enones and en-1,4-diones was developed in the Zezschwitz group at the university of Marburg. In this thesis a new [Rh(cod)OMe]2 catalyzed 1,2-adition containing (R)-BINAP and different aluminium organyles to cyclic en-1,4-diones was found.
