dc.description.abstract | Serotonin (oder 5-HT) ist sehr wichtiger
Neurotransmitter, der verschiedene physiologische
Prozesse in des ZNS reguliert. Diese Effekten werden
durch die große Familie spezialer 5-HT Rezeptoren
vermittelt. Mit Ausnahme vom 5-HT3 Rezeptor, alle
anderen 5-HT Rezeptoren gehören zu der Familie von G
Proteine gekoppelten Rezeptoren (GPCR). Die CPGR dienen
als ein biologischer Schalter für das breites Netz von
Signalwegen durch ihre Interaktion mit heterotrimeren
G-Proteine. GPGR unterliegen oft verschiedenen
post-translationalen Modifikationen, die Rezeptor
Aktivitäten und Funktionen modulieren können. Eine von
dieser Modifikationen ist die Anknüpfung von 16 C
Fettsäure Palmitat. Palmitoylierung ist eine
einzigartige Modifikationen weil dies umkehrbar und
regulierbar ist. Palmitoylierung von GPGR ist in die
Regulation von verschiedenen Prozessen wie Lokalierung,
Interaktionen mit den G-Proteinen, sowie
Phosphorylierung und Desensitisierung.In dieser Studie wurde demonstrieren, dass der
5-HT7a Rezeptor palmitoiliert ist. Durch Kombination
zwischen time course und puls-chase Techniken haben
wir gezeigt, dass der [3H]- Palmitate-Einbau im 5-HT7a
Rezeptor durch die Stimulation mit dem Agonisten
dynamisch reguliert wird.Die C-terminalen Cysteinereste Cys404, Cys438 und
Cys441, wurden als potentialen Palmitoylierungsstellen
identifiziert. Dies wurde durch die ortspezifische
Mutagenese nachgewiesen. Darüber hinaus, haben wir
festgestellt, dass die Palmitoylierung nicht nur in den
Rezeptor C-terminus stattfindet. Funktionale Analyse
von acylierungdeffizienten Mutanten hat eine kritische
Rolle der C-terminale Palmitoylierung für die
konstitutive, agonisten-unabhängige Rezeptoraktivität
gezeigt. Wir haben auch ein neuer Mechanismus
vorgeschlagen, der die Regulation der
Rezeptoraktivitäten durch die dynamische
Palmitoylierung von Cys404 steuert.Außerdem konnten wir für den 5-HT7 Rezeptor
zusätzlich das G12-Protein als neue und bis jetzt noch
nicht beschriebene Rezeptor-Partner identifizieren. Die
Aktivierung des 5-HT7 Rezeptors führt zur
G12-vermittelten Stimulation der kleinen GTPasen RhoA
und Cdc42. In neuronalen Zellen löst die durch den
5-HT7 Rezeptor und G12-Protein initiierte, Cdc42- und
RhoA-gesteuerte Signalkaskade Wachstum der Neuriten und
die Abrundung der Zellen aus. Dies lässt vermuten, dass
Serotonin nicht nur als klassischer Neurotransmitter
wirkt, sondern auch in die Regulation der neuronalen
Architektur involviert ist. | de |

dc.description.abstracteng | Serotonin (5-hydroxytryptamine, or 5-HT) is very
important neurotransmitter which regulates a variety of
different physiological processes within the central
Nervous System and at the periphery. Such effects are
mediated through the large family of the specific 5-HT
receptors. With the exception of the 5-HT3 receptor,
all other 5-HT receptors belong to the family of G
protein coupled receptors (GPCRs). Functionally, GPCRs
act as biological switcher of branched network of
signalling pathways by their interactions with
heterotrimeric G-proteins. GPCRs are often subjected to
the different post-translational modifications, which
can modulate their activity and functions. One of such
modifications is an attachment of 16-carbon fatty acid
palmitate. Palmitoylation is unique between the
post-translational modifications, because it is
reversible and can be regulated. Functionally,
palmitoylation of GPCRs can be involved in regulation
of different processes including membrane targeting,
interaction with G-proteins, basal and
agonist-dependent activity as well as phosphorylation
and desensitisation.In the present study we demonstrated that the
5-HT7(a) receptor undergoes post-translational
palmitoylation. By combining both palmitate
incorporation and pulse-chase techniques we found that
[3H]-palmitate incorporation into the 5-HT7(a) receptor
is dynamically changed after stimulation with agonist
in dose-dependent and time-dependent manner. These
effects were receptor-specific and did not represent
metabolic effects.We also determined cysteine residues Cys404, Cys438
and Cys441 located in the carboxyl terminus of the
receptor as potential palmitoylation sites.
Surprisingly, by analysis of cysteine-deficient mutants
we found that substitution of all three C-terminal
cysteine residues did not completely abolish
[3H]-palmitate incorporation, suggesting that the
5-HT7(a) receptor palmitoylation is not restricted to
the carboxyl-terminal domain.Functional analysis revealed the critical role of
the C-terminal palmitoylation for the modulation of
receptor s constitutive activity. We also proposed a
novel mechanism by which dynamic palmitoylation of
proximal cysteine residue Cys404 may regulate both
agonist-promoted and constitutive activity of the
5-HT7(a) receptor.In addition, we demonstrated for the first time that
the serotonin 5-HT7(a) receptor is coupled both
biochemically and functionally with Ga12 subunit of
heterotrimeric G- protein. We further determined that
the 5-HT7(a) receptor-mediated stimulation of Ga12
protein resulted both in RhoA-dependent neurite
retraction and cell rounding as well as in
Cdc42-mediated filopodia formation. These findings
suggest the important role of serotonin in regulation
of neuronal development in addition to its
well-accepted function as neuromodulator. | de |
