dc.description.abstract | Die Leber als zentrales Stoffwechselorgan
und Teil der körpereigenen Immunabwehr, ist untrennbar mit der
Akut-Phase-Reaktion (APR) im Rahmen einer systemischen
Immunreaktion verbunden. Gerade hinsichtlich des Abbaus toxischer
Substanzen, ausgehend von Gewebeschädigungen übernimmt die
Leberzellen eine Schlüsselrolle. Der Melanocortin-4-Rezeptor
(MC4R), als Mediator anti-inflammatorischer Signale, hat einen
hemmenden Einfluss auf die Produktion verschiedener
pro-inflammatorischer Zytokine. Ziel der vorliegenden
Forschungsarbeit war die Untersuchung der Distribution, wie die
Evaluation der pathophysiologischen Bedeutung des MC4R während der
APR in der Leber. Die Induktion der APR erfolgte durch
intramuskuläre Terpentinöl- Injektion (TO) anhand eines
Rattenmodels. Zu definierten Zeitpunkten nach Injektion, wurde aus
Leber- und Hirngewebe, sowie aus Serumproben RNA extrahiert, zudem
wurden Immunfärbungen von Gewebeschnitten durchgeführt und Proteine
isoliert. Der MC4R konnte mittels PCR- und Western Blot-Analysen,
als auch durch Immunfluoreszenz-und Peroxidasefärbung in der
Rattenleber nachgewiesen werden. 4h, 6h und 12h nach Induktion der
APR konnte eine signifikant gesteigerte Proteinexpression des MC4R
im Vergleich zur unbehandelten Kontrolltiergruppe gezeigt werden.
Die Immunhistologie erbrachte den Nachweis des MC4R im Nukleus von
Parenchym- und Nicht-Parenchymzellen der Leber; des Weiteren konnte
ein Shift der Expression vom Zytoplasma in den Zellkern während der
APR beobachtet werden. Ebenso konnte in Kupfferzell- und
Hepatozytenkulturen der Nachweis des MC4R erbracht werden.
Zusammenfassend zeigt die vorliegende Arbeit, dass der MC4R nicht
nur im Gehirn der Ratte, sondern auch in Leberzellen exprimiert
wird. Nach Induktion der APR zeigt sich dessen Expression
gesteigert. Wir gehen davon aus, dass MC4R-vermittelte
Signalstransduktionsweg, über die Limitierung der Synthese von
pro-inflammatorischen Zytokinen während der APR, eine wesentliche
regulatorische Rolle spielen. | de |

dc.description.abstracteng | The liver as a central metabolic organ and
part of the body’s own immune defence is inextricably linked with
the acute phase reaction (APR) of a systemic immune reaction.
Especially with respect to the breakdown of toxic substances
resulting from tissue damage, liver cells play a key role. The
melanocortin-4-receptor (MC4R) as a mediator of anti-inflammatory
signals acts as an inhibiting factor against the production of
inflammatory cytokines. The objective of this research project was
both an examination of the distribution of the MC4R during the APR
in the liver, as well as an evaluation of its pathophysiological
significance therein. In a rat model, an APR was induced by
intramuscular injection of turpentine oil (TO). At defined points
of time after the injection, RNA was extracted from both liver and
brain tissues and serum samples. In addition, immunostaining of
tissue sections was performed and proteins were isolated. The
presence of the MC4R in the rat liver could be established using
PCR and Western Blot analyses as well as immunofluorescence and
peroxidase stainings. In comparison with an untreated cohort of
control animals, a significantly raised protein expression of the
MC4R could be shown at 4 hours, 6 hours and 12 hours after APR
induction. Immunohistology established the presence of the MC4R in
the nucleus of parenchymal and non-parenchymal liver cells.
Furthermore an expression shift from the cytoplasm to the nucleus
was observed. MC4R was also shown to be present in Kupffer cell and
hepatocyte cultures. In summary, the present work demonstrates that
the MC4R is not only expressed in the brain but also in the liver
of rats. Its expression is increased after APR induction. We assume
that MC4R-mediated signal transduction pathways play a significant
regulatory role by limiting the synthesis of pro-inflammatory
cytokines during the APR. | de |
